Be aware that that requires the Mod Staging Folder (also found in Settings) to be on the same drive as the game. I recommend reading through the in-app documentation before proceeding, as Vortex works a lot differently than other mod managers, especially NMM.

I have SKSE64 installed via vortex, and all of my mods are on the same drive as my Skyrim folder. I get a "Mod Deployment is Pending" message when I launch through Vortex, but they still don't work when I press Deploy. Anyway, I mod skyrim SE as well and have been for a few years on here and I have skyrim installed on my C drive but this on my D drive for space reasons.

Mod Manager is a tool for installing mods in an orderly fashion; they give you lists of mods you installs so you can keep track of what's in your game and what not. Vortex is one of them. I however advises you to uninstall Vortex and go for Mod Organizer 2 instead.
